چکیده
This study aimed to investigate the real-time fluctuation of temperature-sensitive gene expression in a Pythium insidiosum growing at human body temperature (37 ºC) when it can cause life-threatening disease, whereas its natural habitat is agricultural water sources with lower temperature. Genes encoding cytochrome oxidase 2 (COX2), β-tubulin (TUB), and chitin synthase subunit (CHS2) were tested for their infection (27 using RT-PCR. Sixteen strains P. insidiosum, representing 3 phylogeographic preferences, analyzed. The subculturing process 27 ºC was repeated least - cycles. results showed that 37 ºC, COX2 expressed 2.5-fold higher than (p = 0.0347) while level TUB CHS2 mRNA disclosed indistinguishable profiles. These suggested these genes suitable be used as housekeeping temperature-susceptible studies. work likely 1st examining levels Thai strain under thermal stress conditions. Follow-up studies may useful provide valuable insight into pathogenesis, diagnostic, or therapeutic targets further investigation.

Pythium insidiosum: an overview.
Pythium insidiosum is an oomycete pathogenic in mammals. The infection occurs mainly in tropical and subtropical areas, particularly in horses, dogs and humans. Infection is acquired through small wounds via contact with water that contains motile zoospores or other propagules (zoospores or hyphae). The disease, though described as emerging has in fact already been described since 1884. Dependi...

Glomerular epithelial cells (GECs) are known to play a key role in maintaining the structure and function of the glomerulus. GEC injury induced by hyperglycemia is present in early-stage diabetic nephropathy (DN), which is the most common cause of renal failure. In an attempt to identify target proteins involved in the pathogenesis of GEC injury at early DN, we performed the proteomic analysis ...

Extracellular and intraneuronal accumulation of amyloid-beta aggregates has been demonstrated to be involved in the pathogenesis of Alzheimer's disease (AD). However, the precise mechanism of amyloid-beta neurotoxicity is not completely understood. Previous studies suggest that binding of amyloid-beta to a number of macromolecules has deleterious effects on cellular functions. Mitochondria were...
متن کاملAnalysis of COX2 mutants reveals cytochrome oxidase subassemblies in yeast.
Cytochrome oxidase catalyses the reduction of oxygen to water. The mitochondrial enzyme contains up to 13 subunits, 11 in yeast, of which three, Cox1p, Cox2p and Cox3p, are mitochondrially encoded. The assembly pathway of this complex is still poorly understood. Its study in yeast has been so far impeded by the rapid turnover of unassembled subunits of the enzyme. In the present study, immunobl...
متن کاملCaspofungin in vitro and in vivo activity against Brazilian Pythium insidiosum strains isolated from animals.
OBJECTIVES The present study evaluated the susceptibility of 27 clinical isolates of Pythium insidiosum to caspofungin in vitro and correlated the results with the therapeutic response in vivo in rabbits with experimental pythiosis. METHODS The macrodilution method was performed in accordance with the CLSI document M38-A technique. Three reading criteria for MICs were adopted: MIC0, MIC1 and ...
